If i had a computer with 256 MB of RAM (which i do) and i wanted to run a program that demanded, say, 512 MB of RAM, would i be able to temporarily make up for that difference by increasing the size of my Memory Paging File? (I'm planning on getting some more RAM some time in the near future once I have dealt with my graphics card.)

Yes, but the performance of the program will not be optimal.
By default Windows is configured to increase/decrease the size of the Virtual Memory (Paging File) as needed.
If possible, following some of the instructions on this site:
http://support.microsoft.com/kb/314482
would help give better performance until you get more RAM
